Chief United States District Judge Keith Watkins sentenced Jones to 200 months in federal prison on the conspiracy to distribute and possess with the intent to distribute crack cocaine and cocaine powder. He further sentenced Jones to the statutory maximum sentence of 120 months for the use of a telecommunication device in furtherance of a murder for hire. Judge Watkins ordered that those two sentences be run concurrently or at the same time. Once Jones is released from prison he will also have to serve a period of five (5) years of supervised release. Jones remains in the custody of the United States Marshals Service pending placement by the Bureau of Prisons.
As detailed in this AFP report, the full \"US Senate Wednesday unanimously approved legislation to reduce 20-year-old sentencing disparities for offenders caught with crack cocaine versus the drug in its powder form.\" Here are more of the basics: The bill, which must still pass the House of Representatives before President Barack Obama can sign it into law, cuts the 100:1 ratio in the amount of powder cocaine versus crack cocaine that trigger the same sentence.
After 24 years on the books, 15 years of trying, 7 Congresses, 10 hearings, three Sentencing Commission reports, and 75,000 defendants sentenced...today, the U.S. Senate voted -- unanimously -- to make crack cocaine penalties fairer.... If the bill that left the Senate on its way to the House today becomes law, it will take 28 grams of crack cocaine to trigger the five-year sentence and 280 grams to hit the 10-year penalty. And people sentenced for simple possession of crack cocaine will no longer be subject to a five-year mandatory minimum. This is a big improvement over current crack sentencing penalties. It could lower sentences for almost 3,000 people each year. However, the bill is not retroactive and would not help anyone who is already in prison serving a crack cocaine sentence. So, after working on this issue for almost as long as FAMM has been in existence, I'm not thrilled that this is all we got.... [G]iven the politics of the day (and the past 15 years) the Senate bill is likely to be the best we can get. To their credit, there were a number of Democrats and Republicans who supported an even better version of this bill, but not enough. It was clear that nothing short of this compromise would actually make it out of the Senate. Without a Senate bill, crack sentencing reform would be dead once again. The bill now goes to the U.S. House of Representatives where it will meet some obstacles. But because the Senate unanimously approved the bill, it increases its chances that it might make it out of the House. We'll be working closely with House members to see if we can improve the bill.... Since 1995, when Congress killed the reform of the crack sentencing guidelines, nearly 75,000 people have received federal crack cocaine sentences. We will not allow another 75,000 to be sentenced at the current unjustifiable levels. To prevent that, however, we will accept some compromises that are hard to swallow. I don't look forward to that, but I won't let the perfect be the enemy of the good. Too many people have already suffered, which is why we will support this imperfect bill.

We thank the reviewers for their constructive and supportive comments, which we have fully addressed in the specific responses. In summary, we have i) improved the clarity of some of the Figures, ii) added more technical details into some sections of the Materials and Methods and to the Results sections where bioinformatic analysis is discussed, iii) added further discussion on possible batch effects, the choice of animal model for our transcriptomic analysis and the potential pitfalls of comparing data derived from multiple platforms, and iv) corrected a transcribing error in the RAW data file. Minor editorial corrections have also been made to improve readability. Finally, as this study is part of a larger program of work (the CRACK IT Virtual Infectious Disease Research project), we have also now included for the readers benefit, a brief description of this project and an indication of related manuscripts that are currently in preparation for publication.
RNA was isolated from tissue/blood samples and amplified via Agilent low-input Quick Amp labelling kit (Agilent Technologies). Amplified RNA was then assayed with Agilent SurePrint G3 mouse GE 8x60k microarray chips that were scanned with an Agilent C Scanner with Surescan High Resolution Technology (Agilent Technologies). The data were normalized using the percentile shift method to the 75 th percentile. Identification of differentially expressed (DE) genes between infected and naïve samples was performed using the Benjamini and Hochberg false discovery rate (FDR) correction for pairwise comparisons of infected vs. naïve samples 44 . Where multiple probes for a single gene were present, the average expression level was used. This analysis was performed with GeneSpring software (version 9; Agilent) as a standard 5% FDR, with the variances assessed by the software for each t test performed. A 2-fold expression criteria (Log2 FC = 1)was then applied to each gene list. For DE analysis in CRACK-IT_1, a single group of naïve mice were killed at d14 for comparison with samples of infected mice taken at d15, d17 and d21 p.i. For CRACK-IT_2, DE analysis was performed using matched naïve samples for each tissue taken at each time point (see Results).
There are some subtle differences in the size distribution over time, and at d42 there is already some killing of parasites within granulomas (as evident from Fig 1A). This is not fully reflected in change in granuloma size, however.
Minor comments: Many abbreviations are not spelled out at their first appearance; for example, DE and GSEA are explained in full at Results section while already used in Methods. Check other abbreviations too, some are not being spelled out at all.
With peak instantaneous luminosities exceeding 7E33, reducing the impact of radiation effects that affect individual hardware components (SEUs) has been a key activity in 2012. Through improvements in the readout software and firmware, the pixels are now able to automatically reconfigure whenever an SEU causes the pixels to go into a permanent BUSY state. In the rare cases when the reconfiguration fails to fix the problem the affected channels are disabled on the fly by operating in a RunningDegraded state. In addition, the maximum time spent in BUSY has been reduced from 2 seconds down to 0.25 seconds, decreasing the total dead-time from the pixel detector to below 1%. On the online monitoring side the pixel DQM can now show the fraction of ROCs that become silent due to SEUs as a function of time in a run.
A continuous effort was made to fight DAQ instability, identifying sources of downtime and looking for fixes or quick recovery. The use of soft-error recovery was successfully commissioned to the most frequently affected component (TIB-2.8.1, aka FED101
